Several improvements were in the determination of vitamin B_<12> in blood with Euglena gracilis. A standard straight line was used for calculation of determined values. An inoculum of depleted culture gave a low blank, but grew vigorously in the presence of vitamin B_<12>. In inoculating an attention was paid so as to be able to add equal amount of inoculum of uniform concentration by renewing pipettes used for inoculation and stirrng with Komagome-pipettes. One ml was the most suitable quantity of toluene to be added in a test tube containing blood to be digested enzymatically. casein hydrolysate to be supplemented should be more than 4 mg per ml of the basal medium, while the addition of 0.1μg/ml of vitamin B_1 was enough. The above improvements resulted in reasonable recoveries of added vitamin B_<12> of 80-140% as compared with 120-313% in the past. The mean vitamin B_<12> value of blood of 20 healthy subjects was 0.386±0.032mμg/ml, ranging from 0.10 to 0.72mμg/ml.
