It has already been shown that glutamic decarboxylase, γ-aminobutyric-α-ketoglutaric transaminase and pyridoxal kinase of the brain are involved in the running fit incited by 2-methyl-4-amino-5-hydroxymethylpyrimidine (OMP). It has also been found that 2-methyl-4- hydroxy-5-hydroxymethyl-pyrimidine suppresses the OMP induced running fit. The effect of this compound the above enzyme systems was therefore examined. In vitro experiments showed that the compound enhanced the activity of glutamic decarboxylase. Animal experiments too, showed an enhancement of brain glutamic decarboxylase activity by administration of 2-methyl-4-hydroxy-5-hydroxymethylpyrimidine and when given together with OMP, the enzyme suppression is lessened so that the decrease in brain γ-aminobutyric acid is less and moreover, takes place at a slower rate.
