From the findings that 2,5-dimethyl-4-amino-pyrimidine produces a running fit in normal mice and that brain glutamic decarboxylase is reduced during the seizure, investigations were conducted on the mechanism of action of this compound. It was shown that this compound is oxidized in the brain to form 2-methyl-4-amino-5-hydroxymethylpyrimidine (OMP) and the running fit was due to the phosphorylated form of OMP produced at this time. The results of toxicity tests suggest that brain pyridoxal kinase is inhibited by the 2,5-dimethyl-4-aminopyrimidine. It was also found that this compound possesses a toxicity totally different from the running fit inciting action.
