The mechanism of the running fit inciting action of 2,5-dimethyl-4-aminopyrimidine (Compound I) was investigated and the following results were obtained : Compound I does not inhibit glutamic decarboxylase and γ-aminobutyric-α-ketoglutaric transaminase activity in experiments using brain homogenate of normal mice but the two enzymes are inhibited in the case of brain homogenate of vitamin B_6 deficient mice. The inhibition is reversed however, by pyridoxal-phosphate. The above results suggested the involvement of a coenzyme and studies with tyrosine apodecarboxylase of bacteria verified this. It was also found that compound I inhibits pyridoxal-kinase.
