• Acetate heat extraction method (Rosenthal) and diret titrometric method of adding serum to media directly were compared. It was demonstrated that if NaCN was added to serum in extraction, the vitamin B_<12> activity obtained was almost doubled and similar to that obtained by direct method. By adding Co^<60>-labeled vitamin B_<12> to serum in vitro, NaCN was found to enhance liberation of bound vitamin B_<12> from serum protein. It was assumed from the data presented that there are at least two kinds of bound from of vitamin B_<12> present in serum with respect to its heat liberation and Lactobacillus leichmannii utilization.
