1. Candida zeylanoides KY 5802 accumulated remarkable amounts of citric acid and d-isocitric acid in its culture broth. Under suitable conditions, the yield of total citric acid, that is, the sum of citric acid and d-isocitric acid, produced from 7.75% n-paraffins reached 150%, but the ratio between citric acid and d-isocitric acid was about fifty-fifty. Another strain, Candida guilliermondii KY 5822,produced total citric acid in a yield of 90 to 100% from n-paraffins, but the percentage of citric acid in the total citric acid was over 85%. 2. The addition of aconitase inhibitors significantly increased the ratio of citric acid to total citric acid, while the deprivation of ferrous ions from the culture medium slightly increased the ratio. On the other hand, the addition of potassium ferrocyanide considerably decreased the ratio of citric acid to total citric acid, suppressing NAD-linked isocitric dehydrogenase (ICDH) activity. These findings indicate that the ratios between both acids are altered according to the activities of these two kinds of enzymes. 3. Two analytical methods to determine d-isocitric acid and citric acid were established ; one is based upon the determination of the calcium ions of the calcium salts of both acids and the other is a gas chromatographic determination of methylesters of the acids. The latter method was able to determine both acids accurately at the same time.
