It is necessary to obtain the filtration characteristics of culture broth used in large scale filtration.Broth treatment conditions and adsorption on ion exchange resin by the filtrate was studied in the case of vacuum filtration of the culture broth of the antibiotic lividomycin.The network of fibrous mycelia and medium components were considered to be principally responsible for the marked resistance of the cake. In this study, speed of filtration can be increased by adjusting the pH to 3.0 and adding 3% Perlite (filter aid) to the culture broth before filtration. Heating of culture broth to 80℃ shows an effect on the speed of filtration above pH 8.0 but not below pH.3.0 in the culture broth. Specific resistance of the cake increased markedly within about 60 sec in the leaf test; surface filtration is considered to be more important than internal filtration of the precoat.The yield of Lividomycin in the filtrate was 100% below pH 3.0 and above pH 8.4; at pH 5-6,it was only 75%. Lividomycin was adsorbed by a cation exchange resin (Amberlite IRC-50) from filtrate at pH 4.5 (filtration : pH 3.0) and then eluted with 0.5-N NH_4OH with satisfactory results.
