Scale-removed terminal winter buds were aseptically excised from current year shoots of 5〜10-year-old seedlings and cultured in vitro. The best shoot formation was obtained in modified MURASHIGE and SKOOG's medium (mMS) with nitrates reduced to half-strengths as a basal medium supplemented with BAP (6-benzylaminopurine) 0.5 mg/l and GA_3 (Gibberellin 3) 0,l mg/l. Fructose and maltose as well as sucrose were effective for shoot formation and growth. The seasonal differences of winter buds on shoot formation were examined, non-dormant buds showed active shoot formation as compared with dormant buds. Rooting from shoots was better when the cut-ends of shoots were pretreated in 10% (w/w) 1-naphthylacetamide mixed with talc powder and then incubated in Pamisu medium for about four months.
