(9)Activation of Defense-Related Enzymes in Japanese Pear by Acibenzolar-S-Methyl

Mechanism of action of the non-fungitoxic benzothiadiazole acibenzolar-S-methyl (ASM) against pear scab caused by Venturia nashicola, was investigated on the susceptible cultivar 'Kousui'. The whole young leaves of potted trees were sprayed twice with ASM or distilled water and inoculated with the pathogen 7 days after the first treatment. ASM treatment resulted in induction of disease resistance and enhanced activity of three defense-related enzymes. Peroxidase activity was enhanced in ASM-treated leaves 48 hr after inoculation and remained higher during the whole duration of the experiment. However phenylalanine ammonia-lyase activity was weakly enhanced and only 7 days after inoculation. Western blot analyses by using an antibody raised against an acidic class III chitinase of cucumber revealed that a 30 kDa protein was highly detected on ASM-treated leaves 1 day after inoculation. The intensity of this protein band remained higher during the 7 days of the experiment conversely to leaves treated with distilled water in which only a transient increase was noticed.
