The purpose of this study was to examine cell morphology changes associated with creatine monohydrate supplementation (Cr) during an acute load phase and with combined periodized resistance training (4d/week, 14 exiercise, split routine) over a 37-day training period. Twenty-one male subjects, with weight training experience, were randomly assigned to one of three experimental groups. The acute load and supplementation group (ALM) used a 5-day Cr load (0.3g・kg-1 d-1) and a 32-day maintenance supplementation (0.03・kg-1 d-1) with training. The acute load only group (AL) employed at 5-day Cr load and 32-day placebo suppkementation while training. The control group performed similar resistance training but ingested a placebo(PL) throughout the study. Biopsies of the vastus lateralis were taken pre (t1), post 5-day acute load (t2) and post 32-day maintenance phase (T3) and analyzed for cross-sectional area (CSA), fiber type, creatine (Cr), phoshocreatine (PCr), total protein and whole body fluid balance. There was a significant increase in cellular [Cr] for all groups over time, with T2 and T3 significantly higher than TI. Cross sectional area of Type I fibers significantly increased in all groups at T2 and T3. Type II fiber subgroup data (IIa, IIb and IIc), when combined significantly increased in cross-sectional area from T1 to T3 in all groups. No significant differences occurred in total cell water, soluble protein content or percentage fiber type between or within any group over time. The results suggest that 5 days of loading and 32 days of Cr supplementation with training does not significantly affect skeletal muscle morphology more so than training alone.
