This study was conducted to evaluate the effect of exercise training on the regulation of pulmonary artery tone in rats. Twnety 8-week-old male Sprague-Dawley rats were housed in cages in groups of three in an environmentally controlled room, and they were fed standard rat chow and water ad-libitum. The rats were randomly assigned to a control (cont) group (n=10) and an exerise-training (ET) group (n=10) after a 1-week familiarization period. The ET group was forced to run on a small animal treadmill with electrical stimulation (Nat-sume, KN-10, Tokyo) for 8 weeks. All rats were anesthetized with pentobarbital (50 mg/kg, i.p.) immediately after following the training period. The pulmonary artery was carefully isolated from the freshly (10 min) dissected hearts. One aetrery of each pair was denuded of the endothelium by gently rubbing the intimal surface with a roughened steel rodb. The pulmonary arteries were attached to a force transducer (LVS-A, Kyowa, Tokyo). The vasoconstriction responses to the vasoactive substances such as norepincphrine (NE) and phenylephrine (PE) were examined in the physiological salt solutions (PSS). The pulmonary arterial constriction in responsess to PE (10-8 - 10-5 M) and to NE (10-8 - 10-6 M) were wasere significantly less in the ET rats than in the control rats (p<0.05). NE-and PE-induced vasoconstrictions with in endothelium-denuded pulmonary arteries in the ET rats was not different from that in the control rats. Accirdingly, the attenuated vasoconstriction by exercise training in the rat pulmonary artery is thought to be significantly influenced by the endothelial cells. This study suggests that the pulmonary artery vasoconstriction is inhibited by exercise training in the ET rats, and it depends on the a-receptor-mediated endothelium-derived relaxing factor (EDRF)/Nitric oxide (NO).
