Production of Cellulases and β-Glucosidase by Shake Culture of Scytalidium lignicola

The production of cellulases and β-glucosidase was investigated in shake cultures of Scytalidium lignicola CD-48. Amongst various soluble and insoluble carbon sources tested, cellulose (1.75%, w/v)showed maximum production of enzymes. The production of cellulases and β-glucosidase was sensitive to the nitrogen source in the medium, the optimum production was seen when peptone (0.2%, w/v) and KNO_3 (0.2%, w/v) were used as the only nitrogen source. The optimum initial pH for the enzyme production was found to be 5.5,but the maximum production of enzymes occurred when the pH during the cultivation of Scytalidium lignicola CD-48 reached around 6. Mycelia grown from spores in cellulose medium when used as the inoculum (10%, v/v) enhanced β-glucosidase production by about 20%. Tween-80 (0.15%, v/v)and KH_2PO_4 (0.4%, w/v) were found to be optimum for the production of enzymes. Incorporation of a lignocellulosic waste like wheat bran in the medium caused about a 67% increase in β-glucosidase production. It was possible to increase the production of C_1 (6.84 fold), C_x (1.7 fold) and filter paper activity (2.45 fold), and β-glucosidase (16.3 fold) by shake culturing Scytalidium lignicola CD-48 in the modified medium. Cellulases produced by Scytalidium lignicola CD-48 are comparable and the amount of β-glucosidase is substantially higher than that produced by other potential fungi. The significance of the results are discussed.
