Mitochondrial oxidation of different tricarboxylic acid (TCA) intermediates by three radiation induced mutants of Aspergillus niger, one 1st step mutant, HB3 and two 2nd step muatnts, 79/20 and 112/40,together with the mother strain, CA16,was studied. The fungi were grown as surface cultures on 16% commercial sucrose medium at 30℃ and pH 3.5. Mitochondria were isolated during active production of citric acid. The oxidation of citrate, isocitrate, succinate (exogenous) and endogenous substrates were studied by a manometric technique. The citrate yields by the different strains and their rates of oxidation of different TCA intermediates were found to follow an inverse relationship, i.e., the high yielding strains oxidised these TCA intermediates and endogenous substrates at a significantly slower rate than the low yielding strains.
