We developed a new method for the preparation of an axenic culture of the giant coenocytic green alga Valonia ventricosa. Aplanospore formation was induced by exposing the organism to cold shock (15℃) for one day. aplanospore was then aseptically isolated with a syringe. The aplanospore grown on ESS_1 agar medium developed into axenic thalli (2 mm in diameter) after 2 months.
