SCP Production from Steam Exploded Hemicellulose Autohydrolysate by Trichosporon cutaneum

Lignocellulosic waste, the fibrous trash from a paper mill, was pretreated by steam explosion. Using hot water extraction, steam-exploded hemicellulose autohydrolysate lipuor (SEHAL) can be obtained which contains 16.0-47.7 g/l of total sugar, depending on the extraction method. The yeast Trichosporon cutaneum 851 was selected to produce SCP from SEHAL for its ability to use the oligomers of xylose directly and grow fast in the medium. After mutation, a mutant, T. cutaneum 851S, was obtained with an increased protein content of 47.0%. Continuous fermentation was carried out with a dilution rate of 0.33 h^<-1>. A cell concentration of 10.5 g/l, a yield of 0.72 g/g total sugar and a cell productivity of 3.0 g/l・h were obtained. Steam-exploded material was also extracted by spent ammonium sulfite liquor (SASL) from the paper mill. The SASL extract was used as a culture medium without any nutrient supplementation. After adaptation on the SASL extract medium, an acclimated strain, T. cutaneum 851S-6B-1,grew well in the all-waste medium and gave a similar result in continuous fermentation, but with less risk of contamination and at a lower medium cost.
