A hybridoma cell line, AFP-27,was cultivated in a stirred tank with continuous perfusion. Microfiltration membrane was used to partially retain the cells. Using off-line measurement of glucose concentration the perfusion rate was increased in steps with increasing cell concentration. The specific rates of growth, glucose consumption, and anitbody production appear to decrease with time. The need for kinetic models for optimization is discussed.
