Saccharification of Cellulosic Wastes by High-Strength Cellulase System of Mix-Shake Cultivated Scytalidium lignicola and Trichoderma longibrachiatum

High-strength cellulase (C_1-72 units/ml, C_X-27 units/ml and β-glucosidase 3.21 units/ml) was obtained by mix-shake cultivation of Scytalidium lignicola and Trichoderma longibrachiatum. Various cellulosic and lignocellulosic wastes were hydrolysed with the culture filtrate. Paper wastes showed only 25-40% saccharificaiton with the individual culture filtrates of S.Lignicola and T.Longibrachiatum, but a marked increase of 130-150% in the saccharification value was achieved with high-strength cellulase. Agricultural wastes showed a high degree of resistance to enzymatic hydrolysis, giving saccharification values of only 10-15% with individual cultuer filtrates, while the mix-cultivated culture filtrate increased the saccharification value by about 150%. Physical and chemical teratments gave still higher saccharificaton. Hammer-milling gave a 15% increase, while among chemical treatments heat and NaOH were found to be superior to HCI, dioxane, Na-hypochlorite and urea. The Hydrolysate produced was rich in glucose and supported the growth of Candida lipolytica maximally, yielding 970 mg%dry wt.containing 367.2 mg% total protein and 87.3 mg% lipid.
