A modified agar-block method was used for rapid large-scale screening of constitutive cellulase-producing mutants from the fungus Trichoderma reesei. A constitutive mutant isolated by this method, named C-5,makes all three enzymes of the cellulase enzyme complex (endo-1,4-β-glucanase, exo-1,4-β-glucanase (cellobiohydrolase and glucohydrolase) and β-glucosidase) on a wide variety of soluble, non-inducing sugars. These are glucose, sucrose, mannose, soluble starch, and sorbose. A mixture of 1% glucose and 1% sorbose gave the maximum filter paper activity (a measure of total cellulase activity) of 1.8 I.U./ml culture filtrate. The significance of constitutive cellulase production in cellulose bioconversion is discussed.
