Penicillin Acylase Activity in Mutants of Escherichia coli Highly Sensitive to Penicillin G

To obtain cells which have high penicillin acylase activity, several strains highly sensitive to penicillin G were isolated from Echerichia coli IFO 13500 with N-methyl-N'-nitro-N-nitrosoguanidine treatment. Most of these cells had higher acylase activity than the parental strain under inducing conditions. AS2 strain (FERM 70275) had about 6-fold higher activity than the parental strain. The minimum inhibitory concentration of penicillin G for AS2 was 61 IU/ml, while it was 450 IU/ml in the parental strain. The apparent Michaelis constant to penicillin G in AS2 intact cell reaction was 5.0 mM, while it was 16.4 mM in parental cells. AS2 and other strains, which had high acylase activity, were assumed to be mutants in which penicillin G passes easily through their outer membranes.
