Pyruvic Acid Production by Schizophyllum commune : Pyruvic Acid Fermentation by Basidiomycetes (I)

It was found that one strain of basidiomycetes, Shizophyllum commune, produced considerable amounts of pyruvic acid from glucose. The production of pyruvic acid by basidiomycetes has never been reported before. The preculture method and cultural conditions for pyruvic acid production by S. commune no. 550 were then investigated. As a preculture method, a shake culture in medium containing glucose, Polypepton, yeast extract and malt extract for 2 days was revealed to be effective. Concerning the cultural conditions, the following results were obtained. The rate of pyruvic acid production was almost the same within a range of medium glucose concentration of 3 to 10% when the inoculum size was 2.5 ml per 50 ml of medium. As a nitrogen source, only Polypepton gave a high pyruvic acid yield, 38% (19 g/l) after 5 days, among 6 kinds of nitrogenous compounds tested. Acid productivity was not influenced by teh volume of medium between 50 and 200ml. Manganese had a stimulative effect on the acid production. Addition of thiamine to the fermentation medium brought a about a decerase in the amount of pyruvic acid produced and an increase in the mycelial weight. The strain could produce pyruvic acid also from fructose and sucrose in yields of 20% (10g/l) and 31% (15.5g/l), respectively.
