The route of malic acid production from accetate and propionate by Paecilomyces varioti AHU 9417 was investigated. The addition of glyoxylate or fumarate to the fermentation medium increased the yield of malic acid. Fumarate by itself was converted into malic acid, but when glyoxylate was used as a sole carbon source, only glycolic acid was detected in the culture broth. Activities of the glyoxylate cycle enzymes increased dramatically through incubation with acetate and propionate. During the incubation, malic and succinic acids together with a small amount of fumaric acid accumulated in the incubation medium. Acetic acid also accumulated transiently in the propionate medium prior to the accumulation of malic and succinic acids. Results obtained with glucose-incubated mycelia were different from those above. Consequently, it was suggested that the glyoxylate cycle plays an important role in malic acid fermentation of both acetate and propionate by P. varioti AHU 9417.
