Some Major Deviations for Biomass Determination by Indirect Method and Estimation Based on Alkali Consumption

Some factors like the variation of the liquid volume, the variation of cellular nitrogen content and the mass of cells taken with the samples during batch cultivation of microorganisms on diesel oil, were considered for the computation of the ratio between cell mass produced and the mass of alkali consumed to maintain constant the pH of the fermentation medium. The results obtained showed that if such ratios are computed with cell concentration instead of cell mass the deviations can be of the order of 27% caused by the variation of the liquid medium volume. Otherwise, the results showed also that those ratios are variable during batch cultivation on diesel oil probably because of the variations on the nitrogen content of microorganisms. The relative difference between the mass of cells measured and the mass of cells caluculated from the alkali consumption curve can be of the order of 63%.
