Cloning and Characterization of a Polygalacturonase Gene Ap2pg1 from Geotrichum candidum Citrus Race Ap2 Pathogenic to Apple Fruit

Geotrichum candidum citrus race, a fungus that causes sour rot in citrus fruits, secretes a polygalacturonase (PG) that may facilitate the disease. In this research, we isolated a PG gene (Ap2pg1) from G. candidum citrus race Ap2 pathogenic to apple as well as citrus fruits. The predicted protein of the Ap2pg1 consists of 366 amino acids with a putative signal peptide of 27 residues. Reverse transcription-polymerase chain reaction detected Ap2pg1 transcripts in both lemon and apple fruit infected by isolate Ap2. Ap2pg1 was not present in nine isolates of the citrus race of G. Candidum that were nonpathogenic to apple fruit. (Received April 17, 2002 ; Accepted July 18, 2002)
