In order to investigate the existence of a cross-reacting antigen between M. tuberculosis and C. diphtheriae, the passive hemagglutination and conditioned hemolysis were carried out with tuberculous patients' sera and with antisera of rabbits immunized with cells of M. tuberculosis or C. diphtheriae using antigens prepared from tuberculin and diphtherial bacilli. The positive results of cross-reaction with diphtherial antigen were seen in 4 out of 11 tuberculous patients' sera and the positive reactions could be removed by absorption with cells of C. diphtheriae. Similarly, 8 out of 10 antituberculous rabbits' sera showed the positive cross-reaction with the diphtherial antigen which could be removed by absorption with cells of C. diphtheriae. The antibody titers were also reduced by absorption with cells of M. tuberculosis. On the other hand, the rabbits antisera against cells of C. diphtheriae were proved to react positively with both diphtherial and tuberculous antigens. But the antibody titers of antidiphtherial rabbits' sera against the diphtherial antigen remained unchanged after absorption with cells of M. tuberculosis, although the cross-reaction against the tuberculous antigen disappeared by the absorption. Thus, it was presumed from these results that there might exist a cross-reacting antigen between M. tuberculosis and C. diphtheriae.
