The Author studied the immunological relationship between cryoglobulin, normal gamma globulin and myeloma globulin. Two cases of so-called idiopathic cryoglobulinemia, twelve cases of symptomatic cryoglobulinemia and three cases of myeloma were studied. For the precise studies of antigenic determinants, antisera were prepared by immunization of rabbits with normal gamma globulin, each one serum from idiopathic cryoglobulinemia and symptomatic one and three sera from different myelomas. Cryoglobulins were purified by repeated cooling, precipitation, centrifugation and dissolving in warm saline. Myeloma globulins were fractionated and eluted by use of starch electrophoresis. These materials were lyophilized and stored in refrigerator. Agar diffusion method by Ouchterlony was used for the analysis of immunological cross reaction. The symptomatic cryoglobulins from patients with subacute endocarditis, lupus erythematosus disseminatus and rheumatoid arthritis showed the same feature as normal gamma globulin against the anti-normal gamma globulin serum by agar diffusion method. By use of an antiserum against a symptomatic cryoglobulin from one patient with subacute endocayditis, normal gamma globulin was found to have the same determinant group to these of this symptomatic cryoglobulin. Cryoglobulin from idiopathic cryoglobulinemia has a cross antigenicity to the normal gamma globulin, further more seems to have much more specific determinant groups other than the normal gamma globulin does have. In three cases of myeloma one of them has the same determinant groups as normal gamma globulin, the other two have a defect in these determinant groups. Whether the specific determinant groups in idiopathic cryoglobulinemia are of a common property in this disease or different in individual case, remained to be studied. It would appear that these abnormal serum proteins even with the same physico-chemical properties by electrophoresis and ultracentrifugation can be differentiated and classified by immunological method. Cryoglobulin from a patient with idiopathic nature which has specific determinant groups other than those of normal gamma globulin can be determined quantitatively by the vertical agar diffusion method by Oudin with satisfactory results.
