Histamine is released from corpuscles of immunized subjects by the application of corresponding antigen. This experiment was aimed to antigen antibody reaction in immediate type allergy utilizing upper phenomenon. In vitro, to the blood from subjects with immediate type allergy corresponding antigen was added. And, released plasma histamine level was compared with by the case of antigen was added to the blood and by not. Plasma histamine level was determined by a modified fluorometric method devised by Noah and Brand. l) Patients with ACTH hypersensitivity (These patients showed positive cutaneous responses to ACTH by the methods of direct skin test and PK reaction): intense release of histamine was observed, but by the cases of normal subjects no release was observed. 2) Asthmatics; by the cases of patients with positive skin test and PK reaction with house dust intense release of histamine was observed, but the cases of patients with negative skin test and of normal subjects no release was observed. In some these normal subjects house dust was able to evoke positive cutaneous response and positive P-K reaction. 3) Subjects with positive cutaneous response to benzyl penicilloyl protein conjugate : by the case of simple hapten no release was observed, by the case of bivalent hapten weak release was observed and by the case of multivalent hapten intense release was observed. 4) Rabbits immunized with HSA, cephalothin and various penicillins: multivalent haptens were able to release histamine. From these experiments this "in vitro histamine release test" confirmed to be was useful to detect antigens and antibodies concerning immediate allergy or atopic disease.
