Light and electron microscopic studies of the local graft versus host reaction (GVHR) in the kidney which inoculated donor spleen cells in subcapsular spaces of F1 hybrid rats were carried out. The animals were sacrified 15 hours, 3, 5, 7, 14, 28, and 35 days after the subcapsular injection. 1. The most prominent reactions were observed in 7 days after the subcapsular injection and they showed cytologically distinctive cellular infiltration. The changes of renal weight coincide with course of lesions. 2. Early stage of the local GVHR consisted of mainly lymphocytes of large or medium sizes and large pyroninophilic cells. Large pyroninophilic cells possess characteristic appearance by light and electron microscope. After 14th days reactions there were seen presence of plasma cells and lymphocytes. Morphological transformation between large pyroninophilic cells and plasma cells was light and electron microscopically noted. 3. Extensive invasion and destruction of the renal parenchyma were found from 7 to 14 days after the injection. Complete repair of renal parenchyma were seen parallel with disappearance of cell infiltration by local GVHR after 30 days the injection. 4. One may justly say that proliferation cells in early stage of local GVHR derived from donor cells. This reactions is considered to represent method of study of immunologically competent cells. 5. Morphological differentiation and origin of immunologically competent cells were discussed.
