Plaque forming cells (PFC) in the popliteal lymph node and thoracic duct of the rabbit, immunized with sheep red blood cell at the foot pad, were counted by direct and indirect agar plaque formation methods, and they were observed morphologically with phase contrast microscope, using a newly-improved agar plaque method on the slide glass. By the direct method, PFC in the thoracic duct and lymph node were at the maximum, 22.4/l0^6 in number 4 days after immunization, and 380-420/l0^6, 9 to 13 days after immunization respectively. The number of PFC was fewer by the indirect method than that by the direct method, at the early stage, but became almost same as that by the direct method at the four weeks after immunization. PFC were classified into lymphogonia, lymphoblast, basophilic mature lymphocyte and plasma cell. By the direct method, lymphogonia increased to 14.3% 6 days after immunization, and decreased thereafter. Plasma cell was low at the early stage, and increased to 36.4% 28 days after immunization. Basophilic mature lymphocyte ranged from 60% to 70% during the course. Each cell type by the indirect method showed same tendency as by the direct method, but plasma cell was rather increased by the former.
