Effects of Intrathecal Nonnarcotic Analgesics on Chronic Tactile Allodynia in Rats: α_2-Agonists versus Somatostatin Analog

座骨神経を結紮して慢性疼痛モデルラットを作成し、α<SUB>2</SUB>交感神経作動薬(チザニジン、クロニジン)およびソマトスタチン誘導体(オクトレオタイド)の髄腔内注入効果を比較した。Von Frei hairを用いて下肢の疼痛回避反応を定量し、投与前疼痛閾値と薬物注入30分後の閾値を測定し、%MPE=(投与後閾値−投与前閾値)™(15g−投与前閾値)×100を算定した。健側疼痛閾値15g以上、患側閾値4.5g未満で、髄腔内投与を行うと、α2作動薬は25ugでMPEが97%以上で一過性両下肢麻痺や鎮静を生じた。10ugでは副作用は無く、MPEは88-96%であった。オクトレオタイドは最大4ug投与でも副作用は無いが、MPEは49-67%で、α<SUB>2</SUB>作動薬よりもallodyniaの鎮痛効果が弱かった。
