ACUTE EFFECTS OF MERCURIC COMPOUNDS ON PREIMPLANTATION MOUSE EMBRYOS IN VITRO

環境汚染物質は次世代に対して,mutagen/teratogenとして作用する可能性が示唆されている.しかし器官形成期以前の着床前期胚に写える影響はほとんど知られていない.そこで培養マウス着床前期胚の増殖能と蛋白合成能に与える影響を,有機水銀(塩化メチル第2水銀,MMC)と無機水銀(塩化第2水銀,MC)を用いて検討した.ICR糸妊娠マウスのDay 4(腟栓=Day 1)に後期胞胚を採取し,各種濃度の水銀を添加し培養した.培養開始24時間後に水銀無添加培養液へ移し,さらに24時間培養し,合計48時間培養した.培養液はmodified BME+10%FCSを使用し,5%CO_2in air37℃で培養した.培養聞始24時問後,48時間後にair drying法(Tarkowski変法)を用いて胞胚構成細胞数の算定を行なつた.同様処理胚にL-[[chemical formula]]methionineをtracerとして,4時間ラべルし,蛋白合成能を測定した.細胞増殖抑制効果に関し,MMC0.1μMはMC20μMに相当した.蛋白合成能抑制効果に関し,MMC0.1μMは培養開始24時間後ではMC2-5μMに,培養開始48時間後ではMC10-20μMに相当した.したがつて,細胞増殖能に関し,MMCはMCの約200倍の抑制効果を持つこと,また蛋白合成能に関し,約20倍から50倍の抑制効果を持つことが明らかになつた.
