The binding of LH and FSH to granulosa cells was studied using maturing follicles of porcine ovaries, and the effect of sex steroids on the binding was examined. The binding was found to be dependent on the size of the follicles from which granulosa cells were harvested; ^<125>I-LH bound more to the cells from large follicles, while ^<125>I-FSH bound more to the cells from small follicles. It was suggested from the competition of unlabeled LH or FSH to ^<125>I-LH or ^<125>I-FSH binding to the granulosa cells that porcine granulosa cells have respective receptor sites for LH and FSH. The sex steroids, DES and progesterone, showed different effects on the binding of LH and FSH to granulosa cells. DES enhanced ^<125>I-LH binding but suppressed ^<125>I-FSH binding, whereas progesterone suppressed both ^<125>I-LH and ^<125>I-FSH binding.
