RGD-CONTAINING PEPTIDES INHIBIT EXPERIMENTAL PERITONEAL SEEDING OF HUMAN OVARIAN CANCER CELLS

合成ペプチド"RGD (Arg-Gly-Asp)"は, in vitroで細胞, 細胞外基質間の接着を阻害することが知られている。RGDによる癌の血行性転移抑制作用については既に報告があるが, 腹腔内播種性転移については未検討である。我々は, 教室で既に樹立したヒト卵巣明細胞腺癌株JHOC-1を用い, RGDによる腹腔内播種性転移抑制作用について基礎的検討を行った。JHOC-1はヌードマウス腹腔内移植により速やかに腹腔内播種性転移巣を形成し, 移植後10週以内に全個体が死亡する。この転移モデルに対して, RGDを含む合成ペプチド"GRGDS (Gly-Arg-Gly-Asp-Ser)"を各種プロトコールに従って腹腔内投与し, ペプチド投与群及び非投与群について生存期間, 体重変化を比較するとともに, 摘出転移巣病変について病理組織学的検討を行った。GRGDS: 2 mg/mouse×12 (腫瘍細胞移植直後より8時間ごと) 投与群では, 非投与群, 他プロトコール投与群に比較して生存期間は有意に延長し, 25% (3/12) においては腫瘤形成をみることなく長期間 (移植後20週以上) の生存を得た。以上の結果より, RGDによる卵巣癌の腹腔内播種性転移抑制効果が確認されるとともに, 腹腔内播種性転移も血行性転移同様にintegrinsを介した細胞接着に依存することが示唆された。
