6. Responses in Plasma Catecholamines, Renin, Angiotensin, and cAMP in Relation with the Increase in Work Load

The present study was done to investigate the relationship between changes of catecholamines, CAMP and plasma renin activity (PRA), angiotensin concentration, and blood pressure in relation with the increase in work load, and also to confirm the role of catecholamines and renin-angiotensin (RA)system during physical exercise. Six healthy male volunteers with a mean age of 24.2 years, not participated in habitual physical training, were put to the treadmill for 20min (9.2min at maximal test)at different work loads (39.2, 65.8, 82.5, 100% V^^・O_2 max). Each work load exercise was done at a different day. Plasma catecholamines(adrenaline : Ad, noradrenaline : Norad), PRA, angiotensin (Ang I, Ang II), cAMP, blood lactate(LA), systolic and diastolic blood pressure (SBP. DBP) were measured before and after exercise. The following results were obtained. (1) The increments (Δ)of plasma Ad, Norad, cAMP and LA concentration after exercise elevated exponentially in proportion to the increase in work load. The concentration of these components increased rapidly when work load attained the 65.8% V^^・O_2max. (2) Above the 65.8%V^^・O_2max intensity, increment of Norad (ΔNorad ) was 10-fold or more in comparison with that of Ad (AAd). (3) Plasma level of Ang 11 elevated linearly in accordance with the intensity of exercise, although the increase in Ang II concentration after 65.8%V^^・O_2max was about the same as that after 39.2% V^^・O_2max exercise. (4) The change of PRA (ΔPRA)after exercise was 0.6, 3.0, 3.4, 1.7 ng/ml/h, at 39.2, 65.8, 82.5, 100% V^^・O_2max exercise, respectively. (5) Plasma Ang II concentration changed in connection with the increases of Ad, and Norad, but no direct correlation was observed between PRA and CA concentrations throughout the experiment. (6) The increase in SBP and decrease in DBP due to exercise was associated with the work intensity. Correlation of plasma Ang II concentration with SBP, and MBP was r = 0.443. 0.457, respectively. There was no direct relationship between DBP and Ang II, and CA concentrations. (7) Coefficient of correlation between ASBP × HR presented as an index of heart work and ΔAd, and ΔNorad concentration was 0.500, and 0.756, respectively, although no effect of CA on alteration in BP was indicated directly. These results suggested that sympathetic neurvous activity increased rapidly above the moderate work intensity that produces lesser rate of energy production because of anaerobic metabolism, renin secretion was inhibited by exhaustive exercise, and the role of elevated RA system related to the increase in CA during exercise was obscure. Changes in plasma CA and cAMP as well as LA concentration in response to work intensity may be recommended as indices of anaerobic threshold, and also show a mechanism of appearance of anaerobic threshold. Because of small amount of plasma ( 50μl) demanded for cAMP determination in comparison with that for LA (200μl of blood), CA ( 1-2 ml of plasma), cAMP estimation may be a first choice to evaluate physical stress from blood biochemistry.
