A preliminary study on the arrangement of epidermal cells and occurrence of dermal appendages on second leaf of rice seedling was carried out with three cultivars-Kinmaze, Norin Mochi No.1 and Te-Tep, produced at 30 c (control), 20℃ (low temperature) and in 0.1 PPM 2, 4-D solution (chemical treatment). Results obtained were summarized as follows. 1) Size, shape and arrangement of epidermal cells were found to be varietal characteristics. 2) Size and shape of the cells differed according to the position of the cells in the same leaf. 3) Short cells of midrib, larger vein and smaller veins were found to convert into long cells which after a short distance turned into short cells. 4) Long cells near the short cell rows were more or less rectangular in shape whereas those in midportion of intercostal zone were lenticular shaped. Kinmaze had comparatively shorter and broader lenticular cells than those of Norin Mochi No.1 and Te-Tep. 5) Number of short cell rows was reduced by low temperature in all the cultivars, specially that in Kinmaze, as a result of which the number of long cell rows were increased in Kinmaze at the same temperature treatment. It was therefore, contended that the occurrence of short cell and long cell rows were interdepended. 6) Low temperature and 2, 4-D treatment caused reduction and increament respectively in the number of stomata in all the cultivars studied. 7) Occurrence of dermal appendages and their distribution throughout the leaf epidermis had been found to be a varietal characteristic. 8) Size of papillae was comparatively smaller on the cells near the costal zone than that on the midportion of the intercostal zone. Adaxial epidermis contained, in general, smaller papillae than that of the abaxial surface. 9) Kinmaze had comparatively bigger papillae than those of Norin Mochi No.1 and Te-Tep. 10) Number of papillae on subsidiary cells of stomata were four in general and the same was increased by low temperature in Kinmaze and Norin Mochi No.1. 11) All the cultivars, Norin Mochi No.1 in particular, contained many prickle hairs on their adaxial epidermis irrespective of treatment. Low temperature increased the number of prickle hair in Kinmaze, and 2, 4-D treatment increased the same in Norin Mochi No.1 and Te-Tep. Leaf margin was characterized by the radial arrangement of prickle hair only. 12) Macro-hair was a varietal characteristic of Te-Tep and was characterized by their high concentration at the leaf base. 13) Results confirmed that the mode of arrangement of epidermal cells and the occurrence of dermal appendages on the second leaf were more or less same with those of top but one leaf.
