Relationship between Serotypes of Bradyrhizobium japonicum and Their Compatibility with Rj-Cultivars for Nodulation

In order to investigate the preference of Rj-cultivars of soybean for Brady-rhizobium japonicum, rhizobia isolated from field-grown soybeans were classified into nodulation-types A, B, and C based on the compatibility with Rj-cultivars, and relationship between nodulation-types and serotypes of rhizobia was examined by agglutination reactions of B. japonicum isolated from the nodules of the Rj_2Rj_3-soybean cultivars with the antisera against somatic antigens of B. japonicum. Twelve antigens of nodulation-type C isolates (compatible with the Rj_2Rj_3-cultivar, but incompatible with the Rj-4-cultivar) and strain USDA-61 were agglutinated with one or two antisera against the nodulation-type C strains, but did not react with antisera against the nodulation-type A strains (compatible with the Rj_2Rj_3- and Rj_4-cultivars). On the other hand, 3 out of 10 antigens of type A isolates reacted only with antisera against type C strains, and 2 of them reacted with both antisera against type A and C strains. Five antigens reacted only with antisera against type A strains. Therefore, it was assumed that the incompatibility of the Rj_4-cultivars with the nodulation-type C strains did not depend only on their most active antigenic determinants. Nodulation-type C strains were divided into two subtypes. One of them was a strict type C, and the antisera against strains belonging to this subtype reacted only with type C strains as Is-34 and -48. the other was a non-strict subtype, and antisera against this subtype of strain reacted with type A strains in addition to type C strains. The characteristics of the non-strict nodulation-type C strain, Is-21 were examined in relation to the culture age of the inoculant. The cultures which were 12 to 24 days old did not nodulate the Rj_4-cultivar, Hill, but those which were less than 8 days and more than 31 days old nodulated Hill effectively. The bacterial cells harvested from cultures ranging from 8 to 23 days of age were not agglutinated with the antiserum against Is-21. These results suggest that the compatibility of the non-strict nodulation-type C strain Is-21 with the Rj_4-cultivar changed with the culture age, that the change in the compatibility may be related to that in the serological characteristics, and the most active antigenic determinant of Is-21 may not be involved in the incompatibility with Rj_4-cultivars because certain rhizobia of nodulation-type A were agglutinated with the antiserum against Is-21 and the compatibility of cultured cells of Is-21 with Rj_4-cultivars was lost simultaneously with the reactivity with Anti-21.
