MEASUREMENT OF DENITRIFICATION PRODUCTS IN SOIL BY THE ACETYLENE INHIBITION METHOD

The acetylene (C_2H_2) inhibition method was applied in combination with the nitrous oxide (N_2O) measuring system reported previously to measure the amount of denitrified nitrogen (N_2). Acetylene of a rate of 30 ml/day must be supplied from the bottom of pot just before gas sampling to complete the acetylene inhibition in wet soil (90% of water holding capacity) packed in 1.5 liter pots. Acetylene gas contained in the sample gas was removed with a Hg(ClO_4)_2 scrubber before N_2O determination because C_2H_2 masked the N_2O peak recorded on the gas chromatogram. Pot experiment was carried out by using this method. Tanasi soil (volcanic ash) treated with NH_4^+ or NO_3^- (305 μg N/g soil) was incubated at 90% of the water holding capacity in the presence and absence of C_2H_2. Glucose was added to the soil at the rate of 0.1% of oven dry soil. water treatments consisted of Dry (gradual drying)- and Wet (no changes in water content)-treatments, and the periods of sampling were 61 and 22 days, respectively. In the NH_4^+ treatment, at first N_2O and later only N_2 evolved from the wet soil. The amounts of denitrification products in the Dry- and Wet-treatments were 0.04 and 0.05% of nitrogen supplied by fertilizer, respectively. The ratio of N_2-N and N_2O-N was 89:11 in the Wet-treatment and 83:17 in the Dry-treatment. Nitrification of added NH_4^+ was inhibited in the presence of C_2H_2. In the NO_3^- treatment, 29.0% of fertilizer nitrogen was denitrified and the ratio of N_2-N and N_2O-N was 8:92 in the Dry-treatment, 77.7% of fertilizer nitrogen was denitrified and the ratio of N_2-N and N_2O-N was 76:24 in the Wet-treatment.
