Effect of Frankia Inoculation on Growth and Nitrogen-Fixing Activity of Myrica rubra Seedlings Prepared Aseptically

In order to produce efficiently young vigorous Myrica rubra seedlings which can be used for the recovery of vegetation in wastelands, etc., we evaluated the effects of phytohormones on the germination of Myrica rubra seeds, the amount of Frankia strain MrI1 required to enhance growth and nitrogen-fixing activities of the seedling, as well as the effect of nitrate supply on the growth and nitrogen-fixing activity of the seedlings. The results were as follows. 1.The seeds in which the endocarps had been removed were successfully sterilized by a conventional aseptic method with 5% sodium hypochlorite for 7 min. Hormonal treatment was essential to induce the germination of the seeds. The most effective phytohormone was gibberellin A_3 (GA_3) and 60 to 80% of the seeds were fully germinated after treatment with a 10^<-3> M solution for 3 min. Although kinetin promoted the germination at the concentration of 10^<-3> M, the effect was less pronounced than that of GA_3. No such effects were detected when 3-indoleacetic acid and abscisic acid were used. 2.To obtain nodulated seedlings with enhanced growth and nitrogen-fixing activities, it was necessary to inculate more than 10 mL of Frankia strain MrI1 (500-fold diluted solution of the cells collected by centrifugation) per pot. 3.Nitrate supply during the initial 2-month period of cultivation decreased both ARA and nodule fresh weight of the 8-month old seedlings. Especially ARA markedly decreased in the seedlings which received 5 and 10 mM nitrate. 4.A highly positive correlation (significance at 1% level) was observed between the shoot fresh weight and the ARA in 8-month old seedlings. The ARAs of 8-month old seedlings with a greater shoot fresh weight were maintained at a level of more than 1.5 μmol C_2H_4 h^<-1> plant^<-1>.
