Induction of new ectopic bone by auto-osteogenesis factors with artificial vessel(e-PTFE membrane)and autologous vessel bundles

I investigated the possibility of making a simple and effective prefabricated vascularized bone graft to reconstruct jaw bone defects in a rat model(8-week-old male Wistar rats)by using artificial vessels(e-PTFE membranes, Tekunograft^[○|R], Tokyo, Japan), autologous vessel bundles, and auto-osteogenesis factors. In order to make prefabricated vascularized bone grafts on ectopic sites in the control group, an autologous vessel bundle(femur artery and vein)was wrapped in an artificial vessel. Moreover, in the first experimental group, auto-cancellous bone was packed in the site, and in the second experimental group, auto-cortical bone chips were placed. The animals were sacrificed 1, 3, 6, 9 and 12 weeks after the surgical procedure and the osteogenetic process was observed by radiography, histological specimens and electron probe X-ray microanalitic examination. No new bone formation was observed in any stage in the control group. However, in the experimental groups, active bone formation progressed daily after the operation. In the early stages, the greatest amount of new bone formation was confirmed in the first group. In conclusion, prefabricated vascularized bone grafts using blood vessel bundles and an artificial vessel(e-PTFE membrane)are very effective and can be applied clinically as a new source of vascularized bone grafts.
