Platelet-Derived Growth Factor-BB and Epidermal Growth Factor as Pulp Capping Medicaments in Rat Incisors

Factors that regulate growth and development, as well as induce wound healing and tissue regeneration, may facilitate odontoblast differentiation and reparative dentin formation in a mechanically exposed dental pulp. We hypothesize that the use of growth factors as pulpol medicaments may limit the inflammatory response and induce reparative dentin deposition of physiologic quality. The objective of this study is to test the application of platelet-derived growth factor-BB with and without epidermal growth factor as pulp capping medicaments. Direct pulp capping procedures were performed on the mandibular incisors of Sprague Dawley rats. Incisors were treated with one of five regimens and analyzed at 2 and 3 weeks following the procedure. Dycal【○!R】 was used as a positive control, a collagen membrane as a carrier control and a procedurc control with only the sealing materials. The incisors were processed for routine histology and examined by light microscopy. Soft tissue responses were graded for levels of inflammation while hard tissue responses were scored according to the completeness and characteristics of reparative dentin bridge formation. The results were analyzed by one-way ANOVA and Tukey-Kramer tests. The collagen membrane and the PDGF-BB/EGF group showed a better reparative dentin bridge formation at week 2 when compared to the procedure control. The Dycal group exhibited a tendency of diffuse intrapulpal calcification, the PDGF-BB group presented fibrous tissue repair while the procedure control showed irregular calcification without a barrier formation. At week 3, significant difference in hard tissue repair was observed between the PDCGF/EGF group and the procedure control. No significant difference in soft tissue response among the treatment groups were detected. These results indicate that PDGF-BB/EGF may stimulate reparative dentin formation in the rat incisor model.
