Osteoinduction of BMP-Chitin Complex

This study was carried out to develop a new implantable biomaterial combining BMP (Bone Morphogenetic Protein) and chitin. BMP was extracted from bovine cortical bones and half purified. A BMP-chitin complex was synthesized and implanted in the muscle pouches of ddy conventional mice. Chitin and BMP alone were also implanted as negative and positive controls, respectively. The osteoinductive activity the of implants were examined roentgenographically, and histological observations were performed at 10, 20 and 30 days after implantation. At 30 days after implantation, the osteoinductive activity of the implants was quantified according to Ca concentration by means of atomic absorption spectroscopy. The results were summarized as follows. 1) The degree of osteoinduction in mice which received BMP-chitin complex were greater than that in mice that received BMP alone. 2) Histological examination showed that the biological compatibility between surrounding tissues and BMP-Chitin or Chitin alone was excellent. 3) The mechanism of BMP release seemed to be due to diffusion through the embedded chitin. 4) The results of the present study suggested that Chitin was an effective carrier of BMP and was potentially useful in the orthodontic treatments.
