Carboxylesterase (CE) of the organophosphorus insecticide resistant cotton aphid, Aphis gossypii Glover, was separated into several isoelectric point (pI) variants by isoelectricfocusing (IEF). Variants of pI 5.83 and pI 5.57, which were not detected in the susceptible clone existed in much greater quantities than the other variants. These variants were, therefore, thought to mainly contribute to resistance. The pI 5.57 variant was successfully isolated by chromatography as a single protein of 52 kDa when analyzed by SDS-PAGE. On the other hand, the pI 5.83 variant, which was also 52 kDa, was unstable and partially changed to other isoelectric point variants, mainly to the pI 5.57 variant. The pI variation, thus, was thought to have arisen through some structural modification of the pI 5.83 variant during the experimental procedure. However, whether or not the variation really occurs in the live insect's body and what kind of structural modification may contribute to this remain unknown. A partial inner amino acid sequence of the pI 5.57 variant was identified as Ser-Asp-Val-Ile-Ser-Asn-Pro-Phe-Leu-Pro.
