Effects of Adjuvant on the Induction of Helper T Cells in Mice

Certain strains of mice produced much lgE when received ovalbumin (OVA) with Alum as adjuvant (OVA-Alum), but when primed with ovalbumin and complete Freund's adjuvant (OVA-CFA), the mice produced little if any IgE. In an examination of the effects of adjuvants, induction of mRNAs for various cytokines in antigen-presenting cells and lymphocytes was assessed by RT-PCR. When mice were primed with OVA-Alum,the levels of total and antigen-specific IgE were higher than those of mice primed with OVA-CFA. The amount of IL-4 mRNA produced by type 2 helper T cells in mesenteric lymph nodes were more abundant in the mice primed with OVA-Alum than in those primed with OVA-CFA. We extracted mRNA from granulation tissues that were formed subcutaneously after injection of adjuvant, and the expression of mRNAs for IL-1α, IL-1β, IL-12, and TGF-β in the tissues were assayed. The granulation tissues from the mice injected with OVA-CFA expressed higher levels of mRNAs for IL-1α and IL-12 than those from the mice injected with OVA-Alum. The results suggested that the secretion of different type of cytokines from antigen presenting cells induced the polarization of helper T cell subsets in mice primed with these adjuvants.
