Marked variations were observed in biliary metabolites of androsterone in male and female Wistar rats. About half of the rats excreted predominantly monoglucuronides into bile, whereas the remaining rats eliminated mainly mono- and disulphates. Males provided more oxygenated steroid metabolites than females. The injected androsterone glucuronide was almost quantitatively recovered in the bile, while the injected androsterone sulphate was extensively metabolized. Subsequent in vitro studies revealed the existence of striking discontinuous variation in hepatic microsomal UDP-glucuronyltransferase activities towards androsterone but not towards testosterone, bilirubin, p-nitrophenol, and phenolphthalein. The rats with a high level of the transferase activity towards androsterone (HG) and the rats with a low level of the transferase activity (LG) were found approximately in the ratio 5 : 4,and the HG to LG specific activity ratio was about 9,irrespective of the sex. Activation of the latent enzyme with Triton X-100 rather amplified the diversity between the HG and LG groups. Thus, the variability of the UDP-glucuronyltransferase activity should be principally responsible for the large variations in the in vivo metabolism of androsterone in Wistar rats. The results of males suggest the existence of variance of UDP-glucuronyltransferase not only for androsterone but for the oxygenated metabolites of androsterone. The variability of the UDP-glucuronyltransferase towards androsterone was also found in male Donryu and Wistar King rats. In contrast, male CF 1 mice, Hartley guinea pigs, Long Evans rats, and Sprague-Dawley rats did not show such diversity.
