The nuclear gene mutants M_6 101, M134 and M249 that accumulate chlorophyllides a and besterified with incompletely reduced alcohol such as geranylgeraniol, dihydrogeranylgeraniol and tetrahydrogeranylgeraniol were obtained from rice plants(Oryza sativa L. Taichung)by the treatment of seeds with methylnitrosourea. Chlorophyll(Chl)-protein complexes of thylakoid membranes isolated from normal and mutants leaves were analyzed by non-denaturing polyacrylamide gelelectrophoresis. A typical separation pattern gave some bands corresponding to the Chl-protein complexes. The Chl-protein complexes organized in thylakoid membranes showed the redox activity of P700, which indicates that the Chls with incompletely reduced alcohols do not affect substantially the functions of Chl-protein complexes. The results suggest that assembly of Chl-protein complexes occurs without the three-step reductions of Chl alcoholside chain from geranylgeraniol to phytol, but the alcohol moiety is required to prevent leaves from chlorosis under natural conditions.
