In order to clarify the health promoting effect of green tea, the effect of green tea and its components on the two protease activities in granulocytes were examined. Both medullasin and cathepsin G in granulocytes have already been shown to enhance human biophylaxis. In this study effect of green tea and its components on the activity of these two serine proteases in granulocytes, medullasin and cathepsin G, was determined. Both proteases increase DNA synthesis of human lymphocytes, enhance human natural killer (NK) cell activity, and stimulate cytokine production from human monocytes. Mice were sacrificed at 4 weeks after various green tea components were given to measure protease activity in mature granulocytes. When green tea leaves were given to mice, both protease activities decreased, however, when given green tea extract, their activities increased. Addition of the remnant if green tea extract to the diet of mice decreased both protease activities. When epigallocatechin gallate was administered to mice, both protease activities in granulocytes increased. These results indicate that certain green tea components enhance biophylactic activity through the increment of both medullasin and cathepsin G activities in gtranulocytes.
