Microbiosensors for the determination of glutamate, γ-amionbutylate, glucose and lactate were constructed by combining hydrogen peroxide electrode with immobilized enzymes. A hydogen peroxide electrode 800μm in diameter consisting of platinum anode and silver/silver chloride cathode was coated with a thin layer of cellulose acetate. Glutamate oxidase, glucose oxidase or lactate oxidase was immobilized onto the cellulose acetate layer by a glutaraldehyde crosslinking method. The response time of the sensors was about 30 sec. The dynamic ranges of the sensors were 0.1〜1.0mM for glutamate, 1.0〜200mg/dl for glucose and 0.01〜14mg/dl for lactate. The glutamate sensor was applied for determining γ-aminobutylate in combination with enzyme gabase. Both glucose and lactate sensors were utilized for the determination of glucose and lactate in saliva samples taken from subjects before and after an anaerobic exercise (loaded by cycle ergometer for 20 min under controlled cardiac beats 140〜160beats/min). The present microbiosesor method offers a rapid, simple and precise procedure for the determination of biocomponents using small amount of sample.
