Purification and some properties of two proteolytic enzymes (proteinasesI and II) from Clostrodium botulinum TYPE A.

Two proteolytic enzymes, designated proteinases I and II, were obtained from Clostridium botulinum type A. The crude proteinase previously reported from our laboratory, was separated into two proteolytic fractions (Proteinases I and II) by chromatography on QAE-Sephadex. Molecular weights of proteinases I and II, were estimated to be about 28000 and 25000,respectively, by gel filtration on Sephadex G-100. By using cellulose acetate electrophoresis, proteinase II gave a single band; however proteinase I gave two bands as did crude proteinase, which is a mixture of proteinases I and II. Proteinases I and II show the same hydrolytic action on casein and on di-and tripeptides and a similar pH optimum; however, they (I & II) are quite different from each other in the following properties; the electrophoretic mobility, the molecular weight, the heat inactivation and the behavior against Zn^<2+>. Aminopeptidase in C. botulinum has been reported by several investigators^<(4,8,9)>. However, little effort has been devoted to purify and characterize proteolytic enzyme in C. botulinum. Recently, DasGupta et al^<(3)> reported that PTA enzyme isolated from C. botulinum type B hydrolyzed BAPNA and TAME known as a synthetic substrate for trypsin. In a previous paper^<(10)>, Sato et al reported the isolation of two enzymes released from C. botulinum type A and some enzymatic properties. One of the enzymes shows esterolytic activity against BAEE (N-benzoyl arginine ethyl ester) and the other shows proteolytic activity against casein. Thereafter, we investigated further purification of the proteolytic enzyme (crude proteinase) released from C. botulinum type A and separated two proteolytic enzymes (designated proteinases I and II). The present report describes the isolation and characterization of the two proteolytic enzymes.
