Lipid Peroxidation induced by tert-Butyl Hydroperoxide in Bovine Elastic Fibers and Depression of Their Sensitivity to Pancreas Elastase

Oxidative damage in elastic fibers of bovine neck ligament induced with tert -butyl hydroperoxide (t-BHP) was investigated. Elastic fibers used in this study included triglyceride, cholesterol, cholesterol, esters, phospholipid, and fatty acids. Total content of these lipids was 2.9 μg/10 mg fibers. The contents of lipid peroxides and carbonyl compounds in elastic fibers treated with t-BHP were increased in comparison with those in the control. This finding indicates the oxidative denaturation of the regions of lipids and proteins in elastic fibers. Some proteins were released from elastic fibers by t-BHP treatment. They included some amounts of hydrophobic amino acids, such as glycine, alanine, and valine. It was thought that elastic fibers were degraded in the area near the cross-linkings and the oiled coils. These observations show the lack of stability and elastic function in the elastic fibers treated with t-BHP. These changes deceased the resistance of elastic fibers to the pancreas elastase. This lowering of the sensitivity to elastase would be related with the actinic elastosis.
