1. Some methods for the determination of tannase activity have so far been reported, but a more rapid and, within certain limits, sufficiently exact method has been wanted. The difficulty of this determination lies in the fact that there is no suitable method for the measurment of small amount of unreacted tannin or produced gallic acid in the reaction mixture. 2. The method suggested by Nicholson and Rhind was investigated, and some modifications were made in the separation of unreacted tannin and in the color development of gallic acid so as to obtain good and reliable results in the determination of tannase activity. 3. To separate unreacted tannin, 5ml of 1% quinine hydrochloride was used, and to precipitate colloidal quinine tannate, 5ml of 2.5% sodium acetate and 2.5% sodium benzoate mixture (Salt solution) was used per 10ml of reaction mixture. 4. Color intensity developed by gallic acid and ferrous sulfate, was intensified and stabilized by the addition of salt solution. 5. Conditions were examined under which the exclusion of unreacted tannin can be combined with the color development of gallic acid to produce reliable results. 6. A sufficiently exact method was shown for the determination of tannase activity.
