Recentry, angiogenetic factors have been demonstrated to enhance the proliferation and migration of endothelial cells in vivo and in vitro. The specific receptor for urokinase-type plasminogen activator (u-PAR) has been examined relation to cell migration and pericellular fibrinolysis. In the present study, the role of the extracellular proteolysis was analyzed in the model of wound healing by measuring fibrinolytic factors in the tissue extracts and tissue fluid from the chamber with or without fibrin gel subcutaneously inplanted to rat dorsum. The growth of tissue cells was evaluated by measurement of thickness and weight of intrachamber tissue. The volume of tissue in the fibrin-containing chamber was significantly higher than that of the empty chamber on day 5 after the operation. The amidolytic activities of both tissue extracts and fluid in the fibrin-containing chamber were higher than those in the empty chamber on day 5 after the operation. A similar tendency was observed in fibrin and type-I collagen degradative activity on zymography. These findings indicated that upregulated proteolytic activity was effective on the destruction of fibrin clot or interstitium. Both u-PA and u-PAR mRNAs were observed in the intrachamber tissue with or without fibrin gel. The u-PA mRNA in the tissue of the empty chamber was almost 2 times higher than that in the fibrin-containing chamber on days 9 and 14. On the other hand, the u-PAR mRNA content in the tissue of the fibrin-containing chamber was higher than that in the empty chamber on days 5 and 9. These findings suggested that the fibrinolytic activity of tissue was enhanced in the presence of fibrin gel, possibly being upregulated between the late phase of inflammation and the granulation tissue formation via u-PA/u-PAR system.
